High Numerical Aperture - High Resolving Power - Oil Condenser:
This microscope is configured for ultra high resolution, high resolving power, high numerical aperture.
This configuration gives the highest resolution and resolving power.
    Oil Immersion Condenser. 1.4 N.A.
Optimized for use with higher power objective lenses.
Want to get better images? Are you using a 00, high N.A. objective lens, but wasting the resolving power by using a low N.A. dry (0.9 N.A.) condenser?.
Obtain the full resolving power of your objective lens.
This can only be done by matching or exceeding the numerical aperture of the objective with the NA of the condenser.
This condenser is designed for use of immersion fluid between the condenser top lens and the bottom of the glass specimen slide.
Yes, oil immersion condensers take more effort. But when you want publishable quality images, why settle for less? And if you want to use the lower 0.9 N.A., why even have expensive objective lenses?.
    Manual Frame:
Completely manual frame. This makes it easier to operate.
The simplicity of the electronics also means there are less things to go wrong. The motorized frames are prone to failure, especially with their motorized focusing.
Motorized frames have all or most of these following functions as motorized. This frame has all of these functions as nonmotorized: focusing, objective nosepiece rotation, cube turret rotation, transmitted light filter rotation, Koehler illumination field diaphragm opening/closing.
These manual frames are a lot harder to find than the motorized.
    Trinocular Head - Photography Capable:
Photo port on top of head.
Capable of 100% light to eyepieces, 100% to photo port, or split (80% photo, 20% eyepieces).
Ability to shut out extraneous light from the eyepieces that may enter the camera optics.
Eyepieces: W-Pl 10x/23 pair with diopters on both.
Rubber Cupping Eye-Guards to help block extraneous ambient light when looking through the eyepieces.
C-mount attachment for camera is included.
DOES NOT INCLUDE CAMERA. Camera is shown in images to demonstrate that one can be connected.
Differential Interference Contrast - DIC Microscopy:
DIC Magnifications: 40x oil. 63x oil. 100x oil.
The very best DIC images are going to come from the Plan-Apochromat that are certified “DIC” and have red lettering. The included 63x and 100x are both certified “DIC”. These are more expensive than the normal Plan-Apochromat.
The mechanical stage is the DIC version capable of rotating. This is important for optimizing the DIC images.
We tested the DIC on all configured magnifications and obtained excellent textbook quality relief type images for each. Everything works as it was designed to.
DIC is a microscopy technique that gives relief-like images, especially with thick specimens. The 3D effect enhances visualizing structures in thick specimens better than phase. Thick specimens normally are not imaged well in phase contrast. This microscope is capable of imaging in relief or pseudo darkfield, depending on how you adjust it.
    Phase Contrast Microscopy:
Phase Contrast Capability. Configured on 100x objective.
Phase objectives are not the best for fluorescence because the phase ring reduces the low lux fluorescing signal. So the Plan-Apochromat 100x is not phase. A separate, cheaper 100x objective is included for obtaining phase.
Capable of other phase contrast magnifications with other objectives not included.
Condenser has phase annuli: Ph2, Ph3.
Obtained proper centering of phase annulus rings. Includes the required tools for centering.
Includes a phase centering telescope “CT” for optimal phase alignment.
Includes a green filter for optimal phase contrast microscopy imaging.
    Fluorescence Microscopy:
X-cite Metal Halide Fluorescence Illumination for collimated light without the tedious bulb alignment as in HBO units.
The x-cite unit has a variable intensity dial.
The bulbs are also long life, rated for 2000 hours. We are including a new bulb.
UV Protection Filter. This is mounted on the front of the frame to protect your eyes from the intense excitation light on the specimen slide.
    Field Iris Diaphragm.
    Aperture Iris Diaphragm.
These Iris diaphragms are the newer versions that can be pulled out of the frame. The older versions have a simple pull rod with the iris built into the frame. Those versions are very difficult to service and frequently are frozen up or hard to move.
    Manual shutter slider.
IR heatshield (epi-illumination light path) is properly installed in the epi port to protect internal optics.
Includes an additional IR filter on the shutter slider for added protection, This is specifically used to reduce photo bleaching of the fluorescent specimen.
    Fluorescence Dichroic Filters:
All dichroic cubes yielded excellent quality fluorescence images.
DAPI. Zeiss Filter Set 02. G365, FT 395, LP 420. No visible wear.
FITC. Zeiss Filter Set 09. BP 450-490. FT 510. LP 515. No visible wear.
TRITC. Zeiss Filter Set 43HE. BP 550/25. FT 570. BP 605/70. No visible wear. This is a high quality set. Zeiss current website price 94.
TxRed - Some visible wear. The resulting image in the eyepiece was a very bright deep cherry red color.
The rotary filter cube module turret has eight spaces. Blank blockers were installed in all empty spaces.
Transmitted Illumination: Brightfield – Phase - Darkfield - DIC:
    Halogen Illumination: 12V-100W.
    Includes two spare bulbs.
    Rotary Turret style light Condenser.
Numerical Aperature. 1.4 N.A. (oil immersion).
Settings for: Ph2, Ph3, Brightfield, DIC.
    Setting for: Darkfield microscopy.
Filter Wheel (Rear): (manually operated). Daylight blue color correction, Green, Neutral Density ND 6.
Filter Wheel (Front): (manually operated). Neutral Density ND 1.5, 6, 25.
Koehler Field Iris Diaphragm. (manually operated).
    3200 K Illumination Setting Button.
    Koehler Illumination was properly achieved.
IR heatshield (transmitted light path) is properly installed to protect internal optics.
Frosted diffuser filter is properly installed.
Has ability to do cross polarization to increase imaging contrast. Includes polarizer and analyzer.
    Objective Lenses:
    10x. A-Plan 10x / 0.25, ∞/-.
    20x. A-Plan 20x / 0.45, ∞/0.17 mm.
    40x. Plan-Neofluar 40x / 1.30, Oil Immersion, ∞/0.17. Use this for DIC.
63x. Plan-Apochromat 63x / 1.40, Oil Immersion, DIC, ∞/0.17. Red lettering / Rated for DIC. Use this for DIC. Zeiss 2012 price list 75.
100x. Plan-Apochromat 100x / 1.40, Oil Immersion, DIC, ∞/0.17. Red lettering / Rated for DIC. Use this for DIC. Zeiss 2012 price list 88.
    100x. Achromat 100x / 1.25, Oil Immersion, ∞/0.17. Ph3. Phase contrast.
This microscope is configured for high power magnification. And as such, the 10x objective is not able to get a full field of view when Koehler illumination is set.
    Zoom Optovar Magnification Changer:
Intermediate piece with a Zoom Optovar lens system for changing the overall magnification.
Optovar lens - zoom magnifications range: 1x - 2.5x (continuously variable).
This is a continuously variable zoom, not the fixed lens version. This version is extremely rare. Even the fixed lens version for this model is difficult to find.
This zoom lens system changes the final viewing magnification. This is useful for increasing magnification when you don’t want to change the objective.
    Examples:
40x oil objective: Zoom 400x – 1000x. The resulting image is good all the way to the highest zoom.
63x oil objective: Zoom 630x – 1575x. The resulting image is good all the way to the highest zoom.
100x oil objective: Zoom 1000x – 2500x. The resulting image is good through much of the zoom. However, the image is not that good towards the higher zoom magnification because it is beyond the limits of optical microscopy.
Since this microscope has a high numerical aperture condenser, and is capable of higher resolving power, the Optovar lenses are more useful. With low NA systems, simply increasing the magnification may give you marginal or even no benefit with actual resolving power. The same concept, while significantly distorted by sellers of cameras, is the same with resolution specifications of cameras. For example, a really high pixel camera doesn’t ensure you are really going to get more useful information. The more important concept is the design of the lens system.
If you’ve been reading the Chinese microscope ads, you probably think you can simply use a 16x eyepiece and get a beautiful 1600x magnification image. That’s not going to happen. The higher power eyepieces cut the field of view way down in order to get their magnification. For example, the field of view might be as low as 12 or 16 mm. Compare that to the 23 mm from the 10x eyepiece on this unit. They are worthless and just a marketing gimmick. You can always see better with the 10x. The Optovar allows you to still use your 10x.
